Gene SummaryHistones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. This structure consists of approximately 146 bp of DNA wrapped around a nucleosome, an octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a replication-dependent histone that is a member of the histone H3 family. Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element. This gene is found in the large histone gene cluster on chromosome 6p22-p21.3. Application

WB

Western blot analysis of lysates from HeLa cells, using Acetyl-Histone H3-K9/K14/K18/K23/K27 Rabbit pAb at 1:5000 dilution. HeLa cells were treated by TSA (1 uM) at 37℃ for 18 hours. Secondary antibody: HRP-conjugated Goat anti-Rabbit IgG (H+L) at 1:10000 dilution. Lysates/proteins: 25ug per lane. Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Basic Kit. Exposure time: 10s.

ICC/IF

Immunofluorescence analysis of HeLa treated with TSA(1uM,4hours) and HeLa cells using Acetyl-Histone H3-K9/K14/K18/K23/K27 Rabbit pAb at a dilution of 1:100 (40x lens). Secondary antibody:Cy3 Goat Anti-Rabbit IgG (H+L) at 1:500 dilution. Blue: DAPI for nuclear staining.
