Lysine malonylation is a recently discovered reversible modification that can regulate protein activity. Sirt5 is a lysine malonylation modifying enzyme that catalyzes the deprotonation of lysine both in vivo and in vitro. Integrating proteomics and biochemical analysis techniques, it has been found that lysine malonylation can occur in both prokaryotic and eukaryotic organisms, regardless of whether they are histones or non histones. Lysine malonylation may provide a new paradigm for studying intracellular regulation such as energy metabolism in various organisms.
