Gene SummaryThe NF-kappa-B (NFKB) complex of proteins is inhibited by I-kappa-B (IKB) proteins, which inactivate NFKB by trapping it in the cytoplasm. Phosphorylation of serine residues on the IKB proteins by IKB kinases marks them for destruction via the ubiquitination pathway, thereby allowing activation and nuclear translocation of the NFKB complex. The protein encoded by this gene is similar to IKB kinases and can mediate NFKB activation in response to certain growth factors. RACK1 targets MAVS to inhibit type I IFN signaling pathway. (A) Immunoblot analysis of MAVS, p-TBK1(S172), TBK1, p-IRF3(S386), and IRF3 in negative control and RACK1-overexpressing BHK-21 cell lines infected with BEFV of 0.1MOI for 24 h. Anti-β-actin was used as an internal reference control. (B) HEK-293 T cells were co-transfected with Luc-IFN-β and pRL-TK plasmids plus the indicated expression plasmids together with control or RACK1 plasmids. At 24 h post-transfection, the luciferase activities were measured with the dual-luciferase assay. (C) Immunoblot analysis of MAVS in negative-control and RACK1-overexpressing MDBK cell lines infected with BEFV of 0.1MOI for 24 h. The mean and standard deviation values were determined from three independent experiments; *, P ≤ 0.05.

IHC

Phospho-TBK1 (Ser172) Antibody at 1/200 staining Human tonsil tissue sections by IHC-P. The tissue was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The tissue was then blocked and incubated with the antibody for 1.5 hours at 22°C. An HRP conjugated goat anti-rabbit antibody was used as the secondary antibody.
