| Gene Summary | This gene is a member of the ecto-nucleotide pyrophosphatase/phosphodiesterase (ENPP) family. The encoded protein is a type II transmembrane glycoprotein comprising two identical disulfide-bonded subunits. This protein has broad specificity and cleaves a variety of substrates, including phosphodiester bonds of nucleotides and nucleotide sugars and pyrophosphate bonds of nucleotides and nucleotide sugars. This protein may function to hydrolyze nucleoside 5' triphosphates to their corresponding monophosphates and may also hydrolyze diadenosine polyphosphates. Mutations in this gene have been associated with 'idiopathic' infantile arterial calcification, ossification of the posterior longitudinal ligament of the spine (OPLL), and insulin resistance. Western blot analysis of ENPP1 using anti-ENPP1 antibody. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. Lane 1: HEPG2 whole cell lysates, Lane 2: HCCP tissue lysates. After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-ENPP1 antigen affinity purified polyclonal antibody at a dilution of 1:1000 and probed with a goat anti-rabbit IgG-HRP secondary antibody. The signal is developed using ECL Plus Western Blotting Substrate. A specific band was detected for ENPP1 at approximately 150 kDa. The expected band size for ENPP1 is at 105 kDa.
IHC
IHC analysis of ENPP1 using anti-ENPP1 antibody. ENPP1 was detected in a paraffin-embedded section of human endometrial cancer tissue. Biotinylated goat anti-rabbit IgG was used as secondary antibody. The tissue section was incubated with rabbit anti-ENPP1 Antibody at a dilution of 1:200 and developed using Strepavidin-Biotin-Complex (SABC)with DAB as the chromogen.

Flow Cytometry analysis of HepG2 cells using anti-ENPP1 antibody. Overlay histogram showing HepG2 cells stained with Blue line. To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-ENPP1 Antibody at 1:100 dilution for 30 min at 20°C. DyLight®488 conjugated goat anti-rabbit IgG was used as secondary antibody at 1:100 dilution for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG at 1:100 dilution used under the same conditions.
