Gene Full NameTATA-box binding protein
Gene SummaryInitiation of transcription by RNA polymerase II requires the activities of more than 70 polypeptides. The protein that coordinates these activities is transcription factor IID (TFIID), which binds to the core promoter to position the polymerase properly, serves as the scaffold for assembly of the remainder of the transcription complex, and acts as a channel for regulatory signals. TFIID is composed of the TATA-binding protein (TBP) and a group of evolutionarily conserved proteins known as TBP-associated factors or TAFs. TAFs may participate in basal transcription, serve as coactivators, function in promoter recognition or modify general transcription factors (GTFs) to facilitate complex assembly and transcription initiation. This gene encodes TBP, the TATA-binding protein. A distinctive feature of TBP is a long string of glutamines in the N-terminus. This region of the protein modulates the DNA binding activity of the C terminus, and modulation of DNA binding affects the rate of transcription complex formation and initiation of transcription. The number of CAG repeats encoding the polyglutamine tract is usually 25-42, and expansion of the number of repeats to 45-66 increases the length of the polyglutamine string and is associated with spinocerebellar ataxia 17, a neurodegenerative disorder classified as a polyglutamine disease. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2016]

Application

WB

Western blot analysis of TATA binding protein TBP on different lysates with Rabbit anti-TATA binding protein TBP antibody at 1/1,000 dilution. Lane 1: Hela cell lysate (20 µg/Lane) Lane 2: HCT116 cell lysate (20 µg/Lane) Lane 3: 293T cell lysate (20 µg/Lane) Lane 4: HepG2 cell lysate (20 µg/Lane) Lane 5: MCF-7 cell lysate (20 µg/Lane) Lane 6: Jurkat cell lysate (20 µg/Lane) Lane 7: C2C12 cell lysate (20 µg/Lane) Lane 8: NIH/3T3 cell lysate (20 µg/Lane) Lane 9: RAW264.7 cell lysate (20 µg/Lane) Lane 10: Mouse testis tissue lysate (40 µg/Lane) Lane 11: Rat testis tissue lysate (40 µg/Lane) Exposure time: 3 minutes; 12% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/1,000 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:300,000 dilution was used for 1 hour at room temperature.

IHC

Immunohistochemical analysis of paraffin-embedded human bladder carcinoma tissue with Rabbit anti-TATA binding protein TBP antibody at 1/2,000 dilution. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 2 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/2,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

IHC

Immunohistochemical analysis of paraffin-embedded human colon tissue with Rabbit anti-TATA binding protein TBP antibody at 1/2,000 dilution.
