| Gene Summary | This gene encodes a protein which, with 5-lipoxygenase, is required for leukotriene synthesis. Leukotrienes are arachidonic acid metabolites which have been implicated in various types of inflammatory responses, including asthma, arthritis and psoriasis. This protein localizes to the plasma membrane. Inhibitors of its function impede translocation of 5-lipoxygenase from the cytoplasm to the cell membrane and inhibit 5-lipoxygenase activation. Western blot analysis of FLAP on different lysates with Rabbit anti-FLAP antibody at 1/1,000 dilution. Lane 1: U-937 cell lysate, Lane 2: THP-1 cell lysate, Lane 3: THP-1 treated with 50nM TPA overnight then add 5μg/mL LPS for 8 hours cell lysate. Lysates/proteins at 20 µg/Lane. Exposure time: 2 minutes; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/1,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.| Application Notes | WB:1:1000 |
