Gene SummaryThis gene encodes a member of the vascular endothelial growth factor receptor (VEGFR) family. VEGFR family members are receptor tyrosine kinases (RTKs) which contain an extracellular ligand-binding region with seven immunoglobulin (Ig)-like domains, a transmembrane segment, and a tyrosine kinase (TK) domain within the cytoplasmic domain. This protein binds to VEGFR-A, VEGFR-B and placental growth factor and plays an important role in angiogenesis and vasculogenesis. Expression of this receptor is found in vascular endothelial cells, placental trophoblast cells and peripheral blood monocytes. Multiple transcript variants encoding different isoforms have been found for this gene. Isoforms include a full-length transmembrane receptor isoform and shortened, soluble isoforms. The soluble isoforms are associated with the onset of pre-eclampsia.[provided by RefSeq, May 2009]

Application

WB

Western blot analysis of VEGF Receptor 1 on different lysates with Rabbit anti-VEGF Receptor 1 antibody at 1/5,000 dilution. Lane 1: MCF7 cell lysate (20 µg/Lane) Lane 2: Human brain tissue lysate (20 µg/Lane) Lane 3: Mouse brain tissue lysate (20 µg/Lane) Lane 4: Rat brain tissue lysate (20 µg/Lane) Exposure time: 24 seconds; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/5,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:50,000 dilution was used for 1 hour at room temperature.

IHC

Immunohistochemical analysis of paraffin-embedded mouse placenta tissue using anti-VEGF Receptor 1 antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes.The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (1/50) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

ICC/IF

ICC staining of VEGF Receptor 1 in SHG-44 cells (green). Formalin fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 10 minutes at room temperature and blocked with 10% negative goat serum for 15 minutes at room temperature. Cells were probed with the primary antibody (1/50) for 1 hour at room temperature, washed with PBS. Alexa Fluor®488 conjugate-Goat anti-Rabbit IgG was used as the secondary antibody at 1/1,000 dilution. The nuclear counter stain is DAPI (blue).

FC

Flow cytometric analysis of VEGF Receptor 1 was done on A431 cells. The cells were fixed, permeabilized and stained with the primary antibody (1/50) (red). After incubation of the primary antibody at room temperature for an hour, the cells were stained with a Alexa Fluor®488 conjugate-Goat anti-Rabbit IgG Secondary antibody at 1/1,000 dilution for 30 minutes.Unlabelled sample was used as a control (cells without incubation with primary antibody; black).

IP

VEGF Receptor 1 was immunoprecipitated in 0.2mg MCF7 cell lysate with Rabbit anti-VEGF Receptor 1 antibody at 2ug/25ul agarose. Western blot was performed from the immunoprecipitate using Rabbit anti-VEGF Receptor 1 antibody at 1/5,000 dilution. Anti-Rabbit IgG for IP Nano-secondary antibody at 1/5,000 dilution was used for 1 hour at room temperature. Lane 1: MCF7 cell lysate (input). Lane 2: Rabbit IgG instead of Rabbit anti-VEGF Receptor 1 antibody in MCF7 cell lysate. Lane 3: Rabbit anti-VEGF Receptor 1 antibody IP in MCF7 cell lysate. Blocking/Dilution buffer: 5% NFDM/TBST. Exposure time: 24 seconds.
