| Gene Full Name | adhesion G protein-coupled receptor E1 |
| Gene Summary | Predicted to enable G protein-coupled receptor activity. Predicted to be involved in adenylate cyclase-activating G protein-coupled receptor signaling pathway. Predicted to act upstream of or within G protein-coupled receptor signaling pathway and adaptive immune response. Located in external side of plasma membrane. Is expressed in several structures, including cardiovascular system; central nervous system; genitourinary system; hemolymphoid system; and intestine. Orthologous to human ADGRE1 (adhesion G protein-coupled receptor E1). [provided by Alliance of Genome Resources, Apr 2022] |

WB
Western blot analysis of F4/80 on different lysates with Rabbit anti-F4/80 antibody at 1/1,000 dilution. Lane 1: RAW264.7 cell lysate (no heat) (20 µg/Lane) Lane 2: L929 cell lysate (no heat) (negative) (20 µg/Lane) Lane 3: Rat spleen tissue lysate (70℃ heat) (40 µg/Lane) Predicted band size: 102 kDa Observed band size: 160 kDa Exposure time: 2 minutes; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/1,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:100,000 dilution was used for 1 hour at room temperature.
IHC
Immunohistochemical analysis of paraffin-embedded mouse liver tissue with Rabbit anti-F4/80 antibody at 1/1,000 dilution. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
ICC/IF
Immunofluorescence analysis of frozen mouse spleen tissue with Rabbit anti-F4/80 antibody at 1/200 dilution. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for about 2 minutes in microwave oven. The tissues were blocked in 10% negative goat serum for 1 hour at room temperature, washed with PBS, and then probed with the primary antibody (green) at 1/200 dilution overnight at 4 ℃, washed with PBS. Goat Anti-Rabbit IgG H&L (iFluor™ 488) was used as the secondary antibody at 1/1,000 dilution. Nuclei were counterstained with DAPI (blue).| Application Notes | WB:1:1000 IHC:1:200-1:1000 ICC:1:50-1:500 |
