| Gene Summary | This gene encodes amyloid precursor- like protein 2 (APLP2), which is a member of the APP (amyloid precursor protein) family including APP, APLP1 and APLP2. This protein is ubiquitously expressed. It contains heparin-, copper- and zinc- binding domains at the N-terminus, BPTI/Kunitz inhibitor and E2 domains in the middle region, and transmembrane and intracellular domains at the C-terminus. This protein interacts with major histocompatibility complex (MHC) class I molecules. The synergy of this protein and the APP is required to mediate neuromuscular transmission, spatial learning and synaptic plasticity. This protein has been implicated in the pathogenesis of Alzheimer's disease. Multiple alternatively spliced transcript variants encoding different isoforms have been identified. Figure 1. Western blot analysis of anti-APLP2 antibody . The sample well of each lane was loaded with 30 ug of sample under reducing conditions. Lane 1: human SH-SY5Y whole cell lysates, Lane 2: human HepG2 whole cell lysates, Lane 3: human Hela whole cell lysates, Lane 4: rat brain tissue lysates, Lane 5: mouse brain tissue lysates. After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with rabbit anti-APLP2 antigen affinity purified polyclonal antibody and probed with a goat anti-rabbit IgG-HRP secondary antibody. The signal is developed using ECL Plus Western Blotting Substrate. A specific band was detected for APLP2 at approximately 100-110 kDa. The expected band size for APLP2 is at 87 kDa.
IHC
IHC analysis of APLP2 using anti-APLP2 antibody. APLP2 was detected in a paraffin-embedded section of human liver cancer tissue. The tissue section was developed using HRP Conjugated Rabbit IgG Super Vision Assay Kit with DAB as the chromogen.
ICC/IF
IF analysis of APLP2 using anti-APLP2 antibody.APLP2 was detected in an immunocytochemical section of Hela cells. Cy3-conjugated Anti-rabbit IgG Secondary Antibody (red) was used as secondary antibody. The section was counterstained with DAPI (Blue).
FCM
Flow Cytometry analysis of HepG2 cells using anti-APLP2 antibody.Overlay histogram showing HepG2 cells stained with APLP2 Antibody (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-APLP2 Antibody (1:100). DyLight®488 conjugated goat anti-rabbit IgG (1:100) was used as secondary antibody. Isotype control antibody (Green line) was rabbit IgG (1:100) used under the same conditions.
