Poly(dA:dT) Sodium Salt is supplied as a lyophilized powder. To prepare a 1 mg/mL stock solution, reconstitute the powder in 200 μL sterile, endotoxin-free water. Working concentrations and length of treatment can vary depending on the desired effect.
Storage
Store lyophilized at -20ºC, desiccated. In lyophilized form, the chemical is stable for 24 months. Once in solution, store at -20ºC. Aliquot to avoid multiple freeze/thaw cycles.
Background
Poly(dA:dT) Sodium Salt, also known as poly(deoxyadenylic-thymidylic) acid sodium salt, is a double-stranded alternating copolymer DNA model with repetitive synthetic sequences of poly(dA-dT):poly(dT-dA). Poly(dA:dT) Sodium Salt plays a role in the innate immune response by binding to pattern recognition receptors (PRRs), which are used by host cells to detect pathogen-associated molecular patterns (PAMPs) (1). This synthetic B-DNA analog can trigger intracellular DNA sensors, such as DDX41, that leads to stimulator of interferon genes (STING) activation, and downstream activation of the transcription factors NF-κB and IRF-3 (2,3). The effect of cytosolic DNA on the innate immune response is particularly important when studying viral infections (1-3).
